The Gilbert Lab at Louisiana State University investigates how lipoxygenase enzymes recognize, bind, and transform polyunsaturated fatty acids into bioactive lipid mediators. By integrating structural biology, enzyme kinetics, protein biophysics, computational modeling, and AI/ML-guided prediction, we examine how protein dynamics, metal coordination, membrane association, and allosteric regulation control enzyme activity and specificity. Our research spans human, plant, animal, and microbial lipoxygenases, connecting fundamental mechanisms of protein–lipid interactions with broader questions in inflammation, cellular signaling, evolution, and biotechnology. Through this interdisciplinary work, we aim to reveal new principles of membrane-associated catalysis while training students to approach complex biological problems with quantitative, collaborative, and creative thinking.
